摘要
目的初步了解蚕丝支架对雪旺细胞(SchwannCells,SCs)的吸附作用,观察蚕丝支架对SCs生长形态和功能的影响。方法将乳鼠面神经SCs与蚕丝三维支架进行体外复合培养,用倒置显微镜、扫描电镜观察SCs在蚕丝支架上的生长形态、功能及蚕丝支架对SCs的吸附性和组织相容性,并用细胞计数法测定SCs在蚕丝支架上的吸附率。结果倒置显微镜和扫描电镜显示乳鼠面神经雪旺细胞在蚕丝三维支架上的形态和粘附良好,分泌基质旺盛。细胞计数法测定蚕丝对SCs的吸附率为96.325%±0.127。结论蚕丝对SCs具有良好的吸附作用,并能维持SCs的正常形态和功能,蚕丝是SCs立体培养时良好的天然支架。
Objective To understand and observe the attachment, shape and function of Schwann cells ( SCs ) cultured on silk scaffolds in vitro. Methods The SCs of facial nerve in neonatal rats were seeded and cultured in silk threedimentional scaffold, and then the cellular morphology , function, adhesion and biocompatibility with scaffold were evaluated by inverted microscope and scanning electron ( microscope, and the attachment rate of SCs on silk scaffolds was counted. Results The SCs adhered to scaffold well and exhibited the excellent matrix secretion ability under inverted microscope and scanning electronic microscope. The attachment rate of SCs on silk scaffolds was 96. 325%± 0. 127. Conclusion Silks are ideal for SCs attachment, growth and function maintenance , and silks can be used as natural scaffolds for SCs in three- dimensional culture.
