摘要
为探讨丹参治疗溃疡病的分子生物学机制,将72只大白鼠随机分为正常组、丹参组、洛赛克组、模型组。除正常组外,其余各组采用Okabe乙酸涂抹法制作大鼠实验性胃溃疡模型,在造模结束后24h,各组分别以10ml/kg体重给药或给生理盐水,丹参组5g/kg、洛赛克组20mg/kg、模型组与正常组灌服生理盐水,每日1次。在第7、14、21d分别观察溃疡愈合指数、光学显微镜下观察溃疡愈合质量、免疫组化方法检测bFGF、bFGFR的表达情况。结果丹参组可以促进溃疡愈合,其愈合质量优于洛赛克组。溃疡形成后造模各组bFGF、bFGFR表达增加,并且随着溃疡愈合其表达逐渐减弱,在溃疡愈合的各个阶段,丹参组的表达水平均优于其他组别,洛赛克组则与模型组差异不显著。结论丹参能够促进bFGF、bFGFR在溃疡愈合中的表达,这可能是其治疗大鼠乙酸胃溃疡并且提高愈合质量的主要机制。
The aim of this study was to investigate the molecular biological mechanism of Chinese herbal Danshen. 72 rats were randomly divided into four groups. Gastric ulcer were produced in rats by serosal application of acetic acid. Rats were gavaged once daily with lml/100g body weight of: Danshen 5g/kg, Losec 20mg/kg, or normal saline starting 24h after ulcer induction. on day 7,14,21 after treatment, rats were anesthetized with nembutal and stomachs were removed for different detection. We determined(a) Gastric ulcer index ( GUI), (b) histologic assessment of quality of healing ulcer, (c) examined the gastric mucosal expression of bFGF, bFGFR. Results: ( 1 ) Danshen could improve the healing of gastric ulcer. (2) Expression of bFGF ,bFGFR were enhanced after the ulceration, while expression of bFGF ,bFGFR in Danshen group at different phases, were the highest (P〈0.05 versus each group). There were no significant difference showed, compared with model group in losec group at different phase. Conclusion: (1) Danshen could significantly reduce the size of experimental gastric ulcer. (2) restoration of mucosal arshitecture were much better in Danshen group than in losec group. (3) the effect of Danshen on these factors may be crucial for the restoration of mucosal architecture, and may be the basis for the improved quality of mucosal reconstruction by Danshen treatment.
