摘要
目的研究刺梨多糖RRTP-1的理化性质及抗缺氧活性。方法刺梨汁浓缩,乙醇沉淀,得粗多糖。粗多糖脱游离蛋白 质,脱色,DEAE-纤维素柱及Sepharose CL-6B凝胶柱色谱分离纯化得到刺梨多糖RRTP-1。应用凝胶渗透色谱法、气相色谱法 和红外光谱法分别测定其理化性质,并通过观察硫代硫酸钠损伤后各组神经干细胞死亡率和乳酸脱氢酶漏出率,研究其对神 经干细胞硫代硫酸钠损伤的保护作用。结果RRTP-1经凝胶柱色谱和聚丙烯酰胺凝胶电泳证明为相对分子质量分布均一的 多糖,由鼠李糖、阿拉伯糖、未知糖、木糖、甘露糖、半乳糖、葡萄糖和葡萄糖醛酸组成,相对分子质量为3 200。抗缺氧活性实验 表明,加入RRTP-1的实验组神经干细胞损伤有所减轻,高浓度实验组细胞死亡率和乳酸脱氢酶漏出率较低。结论 RRTP-1 对神经干细胞硫代硫酸钠损伤有明显的保护作用。
OBJECTIVE To study the physicochemical properties and anti-hypoxia activity of polysaccharide RRTP-1 from Rosa rox-burghii Tratt.METHODS Crude polysaccharide was obtained from the juice of Rosa roxburghii Tratt by concentration and precipitation with ethanol. After proteins and colouring matter were removed, RRTP-1 was isolated and purified by DEAE-cellulose and Sepharose CL-6B column chromatography from crude polysaccharide and its physicochemical properties were determined by gel permeation chromatography, GC and IR spctrum.RRTP-1 protective effect on neural stem cells was evaluated after the damage induced by Na2S2O3,the cells death rate and LDH leakage were observed. RESULTS The results of column chromatography and electrophoresis illustrated that RRTP-1 was homogeneous and composed of rhamnose,arabinose,unknown sugar, xylose, mannose, galactose, glucose and glucuronic acid, its relative molecular mass was 3 200. The study on anti-hypoxia activity suggested the neural stem cells damage in test groups were alleviated, the cells death rate and LDH leakage in the test group treated with high concentration RRTP-1 were significantly lower than those of Na2S2O3 group. CONCLUSION RRTP-1 owns remarkable protective effect on neural stem cells damage induced by Na2S2O3.
