摘要
目的探讨大明胶囊对2型糖尿病大鼠心肌钾通道KV4.2 mRNA表达的影响。方法 建立2型糖尿病大鼠模型,筛选 空腹血糖值大于16.7 mmol·L-1的大鼠随机分组:大明胶囊大(187.5 mg·kg-1·d-1)、中(93.8 mg·kg-1·d-1)、小(46.9 mg·kg-1 d-1)剂量组、糖尿病模型组,连续用药14 d,用快速血糖仪测空腹血糖值。提取心肌总RNA,采用逆转录聚合酶链式反应(RT- PCR)的方法,观察大明胶囊治疗后2型糖尿病大鼠心肌钾通道KV4.2 mRNA水平的变化。结果2型糖尿病组大鼠心肌钾通 道KV4.2 mRNA表达低于正常组大鼠(P<0.05),经大明胶囊治疗后的心肌钾通道KV4.2 mRNA表达升高(P<0.05),空腹血 糖也明显下降。结论 大明胶囊对2型糖尿病大鼠有明显的降糖作用,并且可以升高2型糖尿病大鼠心肌钾通道KV4.2 mR- NA的表达。
OBJECTIVE To study the effect of Darning capsule on the expression of potassium channel KV4.2 mRNA in the myocardium of the type 2 diabetic rats.METHODS Type 2 diabetic rat model was established. The mrs, whose plasma glucose were higher than 16,7 retool·L^- 1, were divided into 4 groups randomly: high dose group( 187.5 mg·kg^- 1·d^-1 ), middle dose group(93.8 mg· kg^- 1· d^-1 ), low dose group(46.9 mg·kg^-1·d^-1)and control group. The model rats were given with Daming capsule for 14 d.Plasma glucose was determined, The total RNA from the myocardium of rats was measured by reverse transcription polymerase chain reaction (RT-PCR). The effect of Daming capsule on the expression of potassium channel KV4.2 mRNA in the myocardium of the type 2 diabetic rats was observed. RFSLTS The ex- pression of potassium channel KV4.2 mRNA in the myocardium of the type 2 diabetic rats was lower than that in the normal control group( P 〈 0.05), the expression of potassium channel KV4.2 mRNA in the myocardium was increased after being treated with Daming capsule( P 〈 0.05), the values of plasma glucose distinctly decreased. CONCLUSION Darning capsule decreasd the plasma glucose, and increased the expression of potassium channel KV4.2 mRNA in the myocardium of the type 2 diabetic rats.
