摘要 目的建立海金沙藤中咖啡酸的含量测定方法.方法采用反相高效液相色谱法测定,Kromasil 100-5 C18色谱柱(150 mm×4.6 mm),以甲醇-水-冰醋酸-三乙胺(11∶89∶1.5∶0.3)为流动相,检测波长 320 nm,流速1.0 ml·min-1,柱温 35℃.结果咖啡酸与其他成分达到基线分离,线性范围为进样量0.005~0.500 μg(r=0.9999),平均回收率为97.7%,RSD=1.7%.结论所建方法准确、简便,可用于海金沙藤的质量控制. OBJECTIVE To determine caffeic acid in Lygodium japonicum. METHODS An RP- HPLC was performed on a Kromasil 100 - 5 C 18 column ( 150 mm × 4.6 mm), with a mixture of methanol - water - acetic acid - triethylamine ( 11 : 89 : 1.5 : 0.3) as mobile phase . The detective wavelength was set at 320 nm and the flow rate was 1.0 ml·min^- 1. Column temperature was 35 ℃. RESULTS Caffeic acid could be separated from other components completely .The linear range of caffeic acid was 0.005 - 0.500μg( r = 0.9999) and the average recovery of caffeic acid was 97.7 % with RSD of 1.7 %. CONCLUSION This method is accurate, simple and can be used for the quality control of Lygodium japonicum.
