摘要
目的:比较肿瘤来源的热休克蛋白70(HSP 70)与白细胞介素2(IL-2)对荷瘤小鼠肿瘤切除术后的治疗作用。方法:液相色谱法纯化小鼠肿瘤细胞株中的HSP 70,用SDS PAGE及W estern b lot对纯化产物进行定性分析,毛细管电泳鉴定其纯度。动物实验观察HSP 70与IL-2对荷瘤小鼠肿瘤切除术后的治疗作用。结果:单纯手术能延长小鼠的生存期限,HSP 70 10μg组40%小鼠肿瘤最终全部消退,获长期生存(>90 d),平均生存(59.2±29.6)d,与对照组比较差异显著(P<0.05)。手术与HSP 70 10μg联合应用组80%肿瘤完全消退,平均生存(82.0±17.9)d,与对照组、单纯手术组、手术联合应用IL-2 10万u组比较差异显著(P<0.05,P<0.01,P<0.05)。结论:手术能切除局部肿物,但术后易复发、转移,影响长期生存率。HSP 70具有明显全身治疗作用,适当剂量的HSP 70能提高肿物切除后的荷瘤小鼠生存率,比术后应用IL-2治疗效果明显。
Objective: To compare the therapeutic efficacy of heat shock protein 70 (HSP 70) and interleukin-2 (IL-2) in tumor-bearing mice after tumor resection, and to provide evidence for combination therapy of surgery and HSP 70. Methods: HSP 70 was purified from the murine tumor cell line by using liquid chromatography and then identified by using SDS-PAGE and Western blot. The purity was analyzed by using capillary eleetrophoresis. The therapeutic efficacy of HSP 70 and surgery were observed in tumor-bearing mice. Result: Compared with the control group, the life span of tumor-bearing mice was longer in surgery alone group (P 〈 0.05). In 10 lag HSP 70 group, 40% of tumors disappeared, and the mean life span was (59.2 ± 29.6) days. There was significant difference between the 10 lag HSP 70 group and the control group. In combination group of surgery and HSP 70, 80% of tumors disappeared, and the mean life span was (80.2 ± 17.9 ) days. Compared with the control group, surgery alone group, and combination group of surgery and IL-2, the life span was significantly longer in the combination group of surgery and HSP 70 (P 〈 0. 05). Conclusion: HSP 70 has obvious systemic therapeutic effect. Combination therapy of surgery and appropriate dose of HSP 70 can increase the survival rate of tumor-bearing mice after tumor resection, and it is much better than the combination therapy of surgery and IL-2.
