摘要
目的:研究红景天甙对人肝癌细胞系SMMC-7721的诱导分化作用。方法:通过不同剂量红景天甙处理体外培养的SMMC-7721细胞株,采用透射电镜观察细胞亚细胞结构、形态,放射免疫法、酚试剂法分别检测AFP与ALB分泌,重氮试剂法测定γ-GT活力,显微分光光度计定量测定细胞DNA。结果:红景天甙0.5 mg/ml1、.0 mg/ml1、.5 mg/ml均出现细胞增殖抑制,亚细胞结构趋于正常,AFP(alpha feto protein)分泌量下降(P<0.01,vs对照组),ALB分泌量增加(P<0.01,vs对照组),γ-GT(r-glulamgtranserpidage)活力下降(P<0.01,vs对照组),细胞DNA含量降低(P<0.01,vs对照组),并呈剂量效应关系。结论:红景天甙可影响SMMC-7721细胞结构、功能、代谢等指标,诱导其在体外向正常肝细胞分化。
Objective: To study the effect of salidroside on differentiation of SMMC - 7721 cell lines. Methods: By using the method of cell culture exposed to different doses of Salidroside,it was studied the effects of salidroside on SMMC - 7721 cell lines. The ultrastrueture of the cells was observed under transmission deetron microscope,the contents of AFP,ALB,γ - GT,DNA were detected by radioimmunity, phenol reagent, diazo reagent, microspectrophotometer, respectively. Results: The proliferation of treated cells was inhibited by salidroside, the ultrastrocture of the cells tended to normal, alpha feto protein(AFP) was lower, the activity of γ - glutamg - transperpidase (γ - GT) decreased gradually and the DNA content decreased, compared with those of the control cells Conclusion: Salidroside has effects on constitution, function and metabolism of SMMC- 7721 cell line and can induce the cell differentiation tending to normality.
