摘要
目的为寻找高效、价廉、低副作用的糖尿病并发症治疗药物。本实验考察了体外具有较强醛糖还原酶活性的芦丁对大鼠糖尿病肾病的防治作用。方法四氧嘧啶3次皮下注射制备糖尿病大鼠模型,8周后经尿蛋白检测证实糖尿病肾病模型建立。将大鼠随机分成3组:①芦丁治疗组(n=20):糖尿病肾病大鼠每天接受芦丁灌胃200 mg/kg;②糖尿病对照组(DM,n=20):糖尿病肾病大鼠每天接受生理盐水灌胃;③正常对照组(Con,n=20):血糖正常的大鼠每天接受生理盐水灌胃。分别在药物治疗后4、8周检测大鼠的红细胞山梨醇含量、尿量、尿糖、尿蛋白,在药物治疗后8周处死大鼠,检测大鼠肾指数和肾脏LPO。结果芦丁明显改善红细胞山梨醇含量,肾组织LPO、尿量、尿蛋白等指标。结论提示芦丁可改善四氧嘧啶大鼠糖尿病肾病,其机制可能与抑制醛糖还原酶和氧自由基有关。
Objective To study the high effect, low price and side effect drug to treat the complication of diabetes and observe the action to prevention of diabetic nephropathy in vitro. Methods The rat model of diabetes was prepared with injection of alloxan and establishment was confirmed with detection of urine protein after 8 weeks. The rats were divided into three groups randomly. Therapy group was treated with 200 mg/kg lavage for diabetic nephropathy model. Control group was treated with saline lavage for diabetic nephropathy rat and normal group was treated with saline lavage for normal rat. The SBT, urine volume, glucose and protein were detected at 4 and 8 weeks after treatment. The renal index and LPO were detected at 8 weeks. Results The SBT, LPO, urine volume and glucose were improved obviously in therapy group than other groups. Conclusion The results suggested that the rutin could improve the diabetic nephropathy easued by alloxan with related mechanism of inhibit aldose reductase and free radical.
