摘要
目的研究碘酸钾(KIO3)的亚慢性毒性作用。方法采用90 d喂养试验,断乳雌性Wistar大鼠80只,随机分为去离子水对照组和7个剂量组(分别含KIO33 0006、0001、2 0002、4 000、48 000、96 000和192 000μg/L),13周末,作视网膜电图(ERG)检查,处死动物,检测末期血常规、常规生化指标和血清甲状腺激素水平;称量并对脏器进行病理学检查。结果血清TT4、TT3和rT3随染毒剂量的增加而升高,而血清总胆固醇(TC)和甘油三脂(TG)水平从6 000μg/L剂量组即开始显著升高,血白细胞计数显著高于对照组(P<0.05);最高剂量组ERG a、b波振显著低于对照组(P<0.05)。结论碘酸钾的亚慢性毒性试验引起大鼠血清甲状腺激素水平下降、脂质代谢紊乱和视网膜功能性改变。该研究碘酸钾对大鼠的最大无作用剂量为3 000μg/L。血脂为高碘接触的最敏感指标。
Objective To study the subchronic toxicity of potassium iodate (KIO3) and to find its no affected effect level (NOAEL) in rats. Methods 90 day feeding test was adopted. Weanling female Wistar rats were divided into 8 groups randomly. One group drank deionized water as a control group. The other 7 groups drank deionized water containing different doses of KIO3 respectively. At the end of the 13th week, after electroretinogram(ERG) examination on selected randomly rats, all rats were sacrificed. The bloods were collected for testing blood routine, clinical biochemistry index and thyroid hormone. Internal organs were taken carefully for weighing and histopathological examination. Results The levels of serum TT4 , TT3 and rT3 went up with the dose of KIO3 increasing, while total cholesterol(TC) ,triglyeride(TG) in serum and white blood cell (WBC) rose significantly from 6 000 μg/L group( P 〈 0.05). In addition,the amplitude of a and b waves of ERG were significantly lower in the highest KIO3 group rats compared to the control rats( P 〈 0.05). Conclusion KIO3 in 90 day feeding test can result in lower thyroxine in serum,lipid metabolism disorder and retina dysfunction in rats. The NOAEL of KIO3 in rats is 3 000 μg/L. Serum TC level is a sensitive contacting biomarker of excessive iodine.
