摘要
用乙酸和葡萄糖作为诱导试剂,研究酿酒酵母(Saccharomyces cerevisiae)凋亡细胞的毛细管电泳特征及凋亡细胞DNA的荧光光谱特征。发现不同的诱导条件,凋亡细胞的形态、DNA片段化特征不同。荧光光谱证实:乙酸能与脱氧核糖核酸链上的特异位点相结合,形成一种新的加合物,从而断裂DNA;而葡萄糖诱导产生的凋亡过程是一种新陈代谢失调促凋过程,发现两者启动细胞凋亡程序的因素不同。
A programmed cell death(PCD)process induced in yeast Saccharomyces cerevisiae by 800mmol/L acetic acid and 2% glucose respectively was studied with capillary electrophoresis. In capillary zone electrophoresis for the apoptosis yeast cell, it was found that after 6h of treatment with 800mmol/L acetic acid,there were multiple-absorption peaks at 190nm and after 9h of treatment with 2% glucose, there were multiple - absorption peaks at 204nm. For analysis of deoxyribonucleic acid (DNA) flagments in capillary gel electrophoresis, the size of DNA fragments, which induced with acetic acid and with glucose, were about 400bp and 1 600 bp,respectively. In order to investigate the process of DNA fragmentation, the fluorescence experiments for DNA adduct with acetic acid and with glucose were performed, respectively, and the experimental data confirmed that under the different induced condition, the action of the trigger apoptosis was not same.
