摘要
目的 建立测定药物依赖症患者血清中氯硝安定(CZP)浓度的RP—HPLC法,为临床用药提供依据。方法 采用反相高效液相色谱法,色谱柱YWO C18;柱温30℃;流动相为甲醇-水-四氢呋喃=46:53:1;流速1 mL·min-1;波长254 nm。结果 10~1 000 ng·mL-1范围内呈线性,血清中最低检测限5 ng·mL-1,绝对回收 率91.8%-94.8%。10例口服2 mg的患者测1 h时的血药浓度,均在有效血药浓度范围内。结论 用此方法检测 CZP血中药物浓度快速准确,适合于体内CZP的监测及药物动力学研究。
OBJECTIVE To develop a RP-HPLC method for the determination of clonazepam (CZP) in serum of drug-dependent patients to provide a scientific basis for clinical use of CZP. METHODS YWG C18 was used as the reversed column; column temperature was 30 ℃. The mobile phase: methanol-water-tetrahydrofuran= 46 : 53 : 1. The flow rate was 1 mL · min^-1 ; an ultraviolet detector was at 254 nm. RESULTS The linear relation was in the range of 10-1000 ng · mL^-1. The lower detection limit of CZP in the serum was 5 ng · mL^-1. The absolute recoveries of the 4 concentrations of CZP in the serum were 91.8 % - 94. 8 % . The concentrations of the clonazepam (CZP) in the serum of the 10 drug-dependent patients were within therapeutic serum concentration range. CONCLUSIONS This paper provides a rapid, accurate method for monitoring CZP and pharmacokinetic study.
