摘要 目的:对人工种植杜鹃兰的化学成分进行分离和鉴定.方法:采用硅胶、Sephadex LH-20凝胶、ODS柱等色谱分离手段对栽培杜鹃兰的化学成分进行分离、纯化,利用薄层色谱比较及多种波谱方法进行结构鉴定.结果:从杜鹃兰乙醇提取物的醋酸乙酯萃取部分中分离得到8个化合物,鉴定为cirrhopetalanthrin(Ⅰ),7-羟基-4-甲氧基菲-2-O-β-D-葡萄糖(7-hydroxy-4-methoxy-phenanthrene-2-O-β-D-glucoside,Ⅱ),4-(2-羟乙基)-2-甲氧基苯-1-O-β-D-吡喃葡萄糖[4-(2-hydroxyethyl)-2-methoxyphenyl-β-D-glucopyranoside,Ⅲ],对羟基苯乙醇8-O-β-D-比喃葡萄糖(tyrosol 8-O-β-D-glucopyranoside,Ⅳ)vanilloloside(Ⅴ),对羟基苯甲醛(p-hydroxybenzaldehyde,Ⅵ),蔗糖(sucrose,Ⅶ),腺苷(adenosine,Ⅷ).结论:化合物Ⅱ~Ⅷ为首次从该植物中分离得到.cirrhopetalanthrin(Ⅰ)对结肠癌(HCT-8),肝癌(Bel7402),胃癌(BGC-823),肺癌(A549),乳腺癌(MCF-7)和卵巢癌(A2780)细胞表现出非选择性中等强度细胞毒活性,IC50在8.37~13.22μmol·L-1;其他化合物无明显毒性. Objective: To investigate the chemical constituents from the tuber of the planted Cremastra appendiculata. Method: The compounds were isolated by column chromatography over silica gel, Sephadex LH-20 and RP-HPLC, and their structures were elucidated on the basis of spectroscopic analysis. Result: Eight compounds were isolated, and identified as cirrhopetalanthrin ( Ⅰ ), 7-hydroxy-4-methoxyphenanthrene-2- O-β- D-glucoside ( Ⅱ), 4- (2-hydroxyethyl) -2-methoxyphenyl- 1- O-β- D-glucopyranoside (Ⅲ), tyrosol 8- O-β-D-gluco-pyranoside (Ⅳ), vanilloloside (Ⅴ), p-hydroxybenzaldehyde (Ⅵ), sucrose ( Ⅶ), adenosine (Ⅷ). Conclusion: These compounds are isolated from this plant for the first time. All compounds were evaluated against human colon cancer (HCT-8), human hepatoma (Bel7402), human stomach cancer(BGC-823), human lung adenocarcinoma (A549), human breast cancer (MCF-7), and human ovarian cancer (A2780) cell lines, and cirrhopetalanthrin ( Ⅰ) showed non-selective moderate cytotoxicity with IC50 values of 8.4-13.3μmol·L^-1, and other compounds were inactive.
