不同免疫方案对日本血吸虫22.6kDa重组蛋白保护性免疫力的影响 被引量:1

摘要 目的比较福氏完全佐剂(FCA)、福氏不完全佐剂(FIA)与氢氧化铝(Al(OH)3)的免疫增强作用,探讨不同免疫方案对日本血吸虫22.6 kDa蛋白保护性免疫力的影响。方法通过基因工程技术获得rSj22.6/26GST重组抗原的大量表达,用亲和层析法制备rSj22.6/26GST融合蛋白并以凝血酶酶切获得纯化重组日本血吸虫22.6 kDa抗原(rSj22.6)。将纯化rSj22.6抗原分别与FCA、 FIA和Al(OH)3混合后,按照不同的免疫程序分别免疫BALB/c小鼠后进行日本血吸虫尾蚴攻击感染试验。用减虫率及减卵率表示免疫保护效果。结果 Western blot结果证实纯化的酶切产物是日本血吸虫22.6 kDa抗原。rSj22.6抗原与不同佐剂的免疫保护性实验结果提示:与PBS对照组相比.rSj22.6与FCA、FIA的协同作用均能诱导较高的成虫减虫率和总体减卵率(P均<0.01); rSj22.6与Al(OH)3的协同作用虽能诱导产生一定的总体减卵率(P<0.01),但未见有统计学意义的成虫减虫率(P>0.05)。各佐剂干预组所诱导的小鼠抗日本血吸虫感染的保护作用差异均无显著性(P均>0.05)。rSj22.6抗原诱导的保护作用在福氏佐剂的辅助下强于在Al(OH)3作用下的保护作用。结论福氏佐剂和Al(OH)3均可在一定程度上增强rSj22.6免疫小鼠的抗感染保护性免疫力,但福氏佐剂的增效作用强于Al(OH)3。 Objective To compare the immuno-enhancing effects among three adjuvants, Freund's complete adjuvant (FCA), Freund's incomplete adjuvant (FIA) and Al(OH)3 in mice vaccinated by purified rSj22.6 kDa antigen. Methods The rSj22.6/Sj26 GST fusion protein was prepared by affinity chromatography by using glutathione Sepharose 4B. The purified rSj22.6 was cleaved easily from the fusion protein with Thrombin. Mice were immunized three times with rSj22.6 antigen plus FCA, FIA or Al(OH)3. The worms and the liver eggs from the mice each challenged with (40±1) Schistosorna japonicurn cercariae after 45 days were counted. Results The purified rSj22.6 kDa antigen was verified by Western blot method. The immune protection trials showed that rSj22.6 antigen plus FCA or FIA induced a relatively higher worm reduction rate and total egg reduction rate compared with PBS control group (P〈0.01); rSj22.6+Al(OH)3 group showed the statistically significant in total egg reduction rate, but not in worm reduction rate. However, no statistically difference appeared in adjuvant control groups. Conclusion Freund's adjuvant and Al(OH)3 could enhance rSj22.6 induced protective immunity in mice to a certain extent. Freund's adjuvant works more strongly than Al(OH)3.
