摘要
目的研究补阳还五汤对脑缺血再灌注大鼠皮质神经细胞凋亡的影响。方法36只SD大鼠随机分为3组,每组12只:①假手术组;②模型对照组;③补阳还五汤组。线栓法制作大鼠脑缺血再灌注模型,脑缺血-再灌注48 h后TTC染色计算脑梗塞体积,TUNEL染色检测神经细胞凋亡,免疫组化方法检测缺血侧皮质Bcl-2/Bax的表达,电镜观察线粒体的变化。结果脑缺血再灌注后缺血侧皮质神经细胞凋亡增多,同时Bcl-2表达减少,Bax表达增多,Bcl-2/Bax比值下降,线粒体肿胀且大多破裂。补阳还五汤干预后,细胞凋亡减少,脑梗塞体积减小,Bcl-2/Bax比值上升,线粒体肿胀减轻。结论补阳还五汤可能通过抑制神经细胞凋亡而减小脑梗塞体积,其机制可能是通过上调Bcl-2/Bax比值,保护线粒体,防止发生线粒体通透性转变。
Objective To explore the influence of Buyang Huanwu decoction on apoptosis in brain cortex of cerebral isehemia and reperfusion rats. Methods Thirty-six normal male SD rats were distributed randomly to 3 groups: ①sham-operation (n= 12);②control (C,n= 12);③Buyang Huanwu Decoction (BRHWD,n= 12).Cerebral ischemia and reperfusion rat models were made using the method of thread inserting right middle cerebral artery occlusion( MCAO). The infarct volume was measured 48 hours after reperfusion. At the same time, the apoptosis was detected by TUNEL staining. The expression of Bcl-2 and Bax in different groups were detected by means of immunohistochemistry( ABC method). The changes of mitochondria were observed with electron microscopy. Results Apoptosis in isehemic brain cortex was increased, expression of Bcl-2 was decreased and Bax was increased. The ratio of Bcl-2/Bax was decreased. In addition, the mitochondria swelled obviously. When the model rats were administered with Buyang Huanwu decoction, the brain infarct volume relieved (p 〈 0.05), apoptosis decreased, the ratio of Bcl-2/Bax increased and mitochondria swelled slightly. Conclusion Buyang Huanwu decoction can relieve infarct volume possibly by decreasing apoptosis in brain cortex. The decoction may play a good part in increasing the ratio of Bcl-2/Bax, protecting mitochondria and preventing mitochondrial permeability transition.
