摘要
目的探讨快速延迟整流钾通道(ether-a-go-go-relatedgene,erg)在CDl小鼠胃肠道的表达及对胃肠道电活动的影响。方法用逆转录聚合酶链反应(RT-PCR)检测erg mRNA在成年CD1小鼠胃肠道的表达;并用细胞外记录法记录和比较erg通道特异性阻滞剂E-4031干预前后离体结肠慢波电位的变化。结果在CDl小鼠胃肠道不同部位均存在着erg mRNA的表达;E-4031干预后,结肠慢波电位频率减慢(P<0.01),时程延长(P<0.01),叠加在慢波电位上的快波电位数量增加,幅度增大,但慢波电位的幅度变化无统计学差异(P>0.05)。结论erg在CDl小鼠胃肠道不同部位表达,ergK+通道在调节胃肠道平滑肌自发性电节律过程中扮演了重要的角色。
Objective To test the expression and possible role of ether-a-go-go-related gene (erg) protein K^+ channels in gastrointestinal tract of CD1 mice. Methods The expression of erg mRNA in the gastrointestinal tract of CD1 mice were tested by RT-PCR. Extracellular voltage recordings was used to compare the slow-wave activity of colon in the absence or presence of E-4031 (1μM, 30min). Results RT- PCR showed the presence of ERG mRNA in the gastrointestinal tract of CD 1 mice. E-4031 markedly increased tissue excitability. In the presence of E-4031 (1μm, 2 min), slow-wave duration increased, slow-wave frequency decreased and the number and amplitude of action potentials superimposing slow-wave increased,but the amplitude of slow-wave didn't change significantly. Conclusion erg K^+ channels contribute to the membrane potential and play a role in modulating spontaneous electrical activity in the gastrointestinal tract.
