摘要
目的探讨茵陈五苓散对动脉粥样硬化(AS)大鼠细胞凋亡的影响。方法采用高脂饲料喂饲法复制大鼠动脉粥样硬化模型,70只雄性Wistar大鼠随机分为正常组、模型组、茵陈五苓散组、胶股蓝组,造模1个月后抽样检测大鼠主动脉,以发现动脉粥样硬化斑块为造模成功指标,经治疗1个月后分别检测血脂、血液流变学,用HE染色观察主动脉组织学变化,用透射电镜观察大鼠主动脉组织超微结构变化,用免疫组化法观察大鼠主动脉血管平滑肌细胞(VSMC)细胞凋亡。结果茵陈五苓散与胶股蓝组比较,能降低TC、TG、LDL,升高HDL,均有非常显著性意义(P<0.01),降低血浆黏度、全血中切、红细胞压积、血小板黏附率具有显著性意义(P<0.05),消退斑块具有显著性意义(P<0.05),维持主动脉组织结构,抑制细胞凋亡具有显著性意义(P<0.01)。结论茵陈五苓散具有良好的抗动脉粥样硬化作用,它可能是通过抑制细胞凋亡来实现的。
Objective To observe the effect of Yinchen wulin powder on apoptosis of rat with atherosclerosis(AS). Method The AS model of rat was made by feeding high grease food. 70 wistar rats were divided into four groups: normal, model, tablet Gypenosides and Yinchen wulin powder group. The level of serum lipid and blood rheology were tested after one month; the change of aorta histology was observed by HE staining; the change of aorta ultrastructure with transmission electronic microscope; the apoptosis of aorta vascular smooth muscle (VSMC)with immunohistochemieal methed. Result In the therapy group, the level of TC, TG, LDL decreased and the level of HDL increased( P 〈 0.01), plasmviscosity, erythrocyte hematocrit and platelet adhesion ratio decreased significantly( P 〈 0.05) ; the histology of aorto didn't change much compared with gypenoside group. Conclusion Yinchen wulin powder has significant effect on AS and the therapeutic molecular mechanism is inhibition of apoptosis.
