摘要
目的:建立测定人血清中胺碘酮的高效液相色谱法.方法:色谱柱:Ultrasphere C18(250 mm×4.6 mm,5μm),流动相:乙腈-甲醇-0.01 mol·L-1醋酸铵液(30:55:15),100 mL中含0.2 mL三乙胺,用冰醋酸调pH值至(4.00±0.05),流速:1.2 mL·min-1,检测波长:240 m,柱温:室温.结果:线性范围0.062 5~8.00mg·L-1,平均回收率为100.9%~107.9%,日内RSD小于7%,日间RSD小于8%.结论:此法可用于胺碘酮的血药浓度监测.
OBJECTIVE To establish a method for determining the concentration of amiodarone in serum by HPLC. METHODS The column was Ultrasphere C18 ,the mobile phase was acetonitrile-methanob0. 01 mol·L^-1 ammonium acetate[100 mL containing 0. 2 mL triethylamine, pH value was adjusted to (4. 00 + 0. 05) by acetic acid] (30:55:15), the detection wavelength was 240 urn, the flow rate was 1.2 mL.·min^-1, the temperature of column was at room temperature. RESULTS The calibration curves were linear in the range 0. 062 5 - 8.00 mg·L^- 1 for amiodarone, the recovery was 100. 9 % - 107. 9 %, inter-day RSD was less than 7% and intra-day RSD was less than 8%. CONCLUSION The method could be used for therapeutic amiodarone monitoring.
