摘要
目的研究FcεRⅠ-β的I181L、V183L和E237G突变在四川地区11岁以下汉族儿童中的存在和频率,判断这些突变与儿童哮喘的相关性。方法利用扩增阻滞突变系统聚合酶链技术(ARMS-PCR)对四川地区的48例汉族哮喘儿童进行FcεRⅠ-β的1811、83、237三个位点的突变进行检测和分析,并与40例健康儿童进行对照。结果在哮喘组中检测到I 181L突变5例(10.4%),V 183L突变2例(4.2%),E 237G突变6例(12.5%),在对照组中未发现I 181L、V 183L、E 237G突变子。结论四川地区汉族儿童中存在I 181L、E 237G突变,不存在V 183L突变或突变率极低。I 181L突变与哮喘存在相关性(P<0.05),E 237G突变和哮喘有相关性(P<0.025),而V 183L与哮喘不存在相关性(P>0.05)。
Objective To detect the mutation frequency of I181L, V183L, E237G in FccR I -β,and assess their association with asthmatic children whose ages were below 11 years and inhabited in Sichuan province. Methods We used amplification refractory mutation system-polymerase chain reaction (ARMS-PCR) to detect and analyze three mutations(I181L, V183L and E237G) in the beta chain of the high affinity IgE receptor (FccR I -β) in 48 asthmatic subjects and 40 healthy controls who came from Sichuan province of China. Results In the asthmatic group, there were 5 samples of I181L mutation (10.4%), 2 samples of V183L mutation (4.2%),and 6 samples of E237G mutation (12.5%). There was no I181L, V183L and E237G mutations in the control group. Conclusion I181L and E237G mutation exists in the children whose ages were below 11 years in Sichuan province of China; while V183L mutation is not found or their frequency of mutation is very low. There is the correlation between asthma and the mutation of I181L (P〈0.05) and E237G (P〈0. 025), but the correlation is not found between asthma and V183L mutation (P〉0.05).
