CTGF反义寡核苷酸对肾小管上皮细胞胶原Ⅲ分泌的影响 被引量:1

Inhibition of Type Ⅲ Collagen Secretion by Connective Tissue Growth Factor Antisense Oligodeoxynucleotides in Tubular Epithelial Cells

摘要 目的观察结缔组织生长因子(CTGF)反义寡核苷酸对TGF-β1诱导的肾小管上皮细胞NRK52E胶原Ⅲ分泌的影响。方法采用脂质体介导的方法将CTGF反义寡核苷酸导入细胞,转化生长因子β1(TGF-β1)刺激48 h后,用RT-PCR、ELISA法观察CTGF和胶原Ⅲ的表达。结果TGF-β1能够诱导NRK52E细胞表达CTGF并进而促进胶原Ⅲ的分泌,CTGF反义寡核苷酸导入细胞后可以大部分取消TGF-β1的作用,而对照组的寡核苷酸没有此作用。结论CTGF反义寡核苷酸能够特异地抑制CTGF的表达,进而阻止细胞外基质的生成,这可能是治疗肾间质纤维化的一条新途径。 Objective To investigate the influence of Connective tissue growth factor(CTGF) atisense oligodeoxynucleotides (AS-ODNS) on type Ⅲ collagen secretion induced by tansforming growth factor-β1 (TGF-β1). Methods CTGF AS-ODNS and control reverse ODNS were transfected into cells by cationic lipofection, cells were stimulated with TGF-β1 for 48h. The expression of CTGF and type Ⅲ collagen was detected by RT-PCR and ELISA. Results TGF-β1 was able to induce the expression of CTGF and further facilitate the secretion of type Ⅲ collagen in cultured NRK-52E cells, CTGF AS-ODNS inhibited the expression of CTGF gene, and down-regulated type Ⅲ collagen expression in response to TGF-β1, but control oligodeoxynucleotides not. Conclusion In cultured NRK-52E cells, CTGF AS-ODNS transfection specifically inhibits TGF-β1-induced CTGF expression and further attenuates type Ⅲ collagen expression. It suggests that blocking CTGF with AS-ODNS could be a possible therapeutic strategy against tubulointerstitial fibrosis.
