摘要
目的:采用高效液相色谱法测定肌苷氯化钠注射液中肌苷的含量及有关物质。方法:采用Hypersil C_(18)柱(10μm,4.6mm×200mm),以0.05mol·L^(-1)醋酸钠缓冲溶液(以1mol·L^(-1)醋酸调pH至5.0),流速1.5·min^(-1),紫外检测器于248nm测定。结果:HPLC法测定的线性范围为0.1~0.8mg·mL^(-1),r=0.9998,最低检测限2.5ng;样品溶液在12h内稳定,日内精密度和日间精密度(RSD<1.0%)良好。回收率为99.93%。结论:采用高效液相色谱法测定肌苷氯化钠注射液中肌苷的含量及有关物质的方法简便,结果可靠。
Objective: To determine the content and the related substances of inosine in inosine and sodium chloride injection by HPLC. Methods: The column: Hypersil C18 column( 10μm, 4.6mm×200mm) ; The mobile phase; buffer solution with 0.05mol·L^-1 Sodium acetic(adjusted to pH5.0 with 1mmol·L^-1 acetic acid) ;The flow rate: 1.5ml·min^-1;The content of inosine and its related substances were deteced at 248nm on the same condition.Results: The linear rarge of the determination by HPLC is 0.1-0.8mg·mL^-1; The correlation coefficient is 0.9998; the least detective range is 2.5ng; the sample solution is stable within 12h; the precision in a day and the precsion in days are good(RSD〈 1.0% ) .The recovery are 99.93% .Conclusion: The method to detemaine inosine the content and the related substances in inosine and sodium chloride injection by HPLC is easy to operate and the results the detemination are accurate.
