摘要
目的观察中药单体青藤碱对体外培养类风湿关节炎患者树突状细胞核转录因子-κB活性的影响。方法分离12例类风湿关节炎患者外周血单核细胞,分为青藤碱高、中、低剂量组和空白对照组。采用GM-CSF,IL-4联合刺激分化,于分化第6天给予青藤碱干预,48 h后于培养第8天收获细胞及培养上清。抽提核蛋白采用凝胶迟滞实验检测核转录因子-κB的活性,酶联免疫吸附实验检测培养上清IL-12的表达。结果与对照组相比,经青藤碱高、中、低剂量干预的树突状细胞核转录因子-κB活性及IL-12表达均降低,不同剂量组之间两者存在正相关。结论青藤碱可能通过抑制树突状细胞转录因子-κB活性,阻断其分化成熟,阻止了它在类风湿关节炎发病中的抗原提呈作用。
Objective To observe the effect of sinomenine on the activity of nuclear transcription factor kappa B in dendritie cells in rheumatoid arthritis in vitro.Methods Peripheral blood mononuclear cells isolated from 12 rheumatoid arthritis patients were divided into four groups: sinomenine high, middle, low concentration groups and control group and were cultured with GM - CSF, IL - 4. The sinomenine group was intervened with different concentration of sinomenine after 6 days and harvested 48 housr later. The activity of nuclear factor kappa B was examined by electrophoretic mobility shift assay and IL - 12 released by dendritic cells was measured by enzyme linked immunosorbent assay. Results Nuclear factor kappa B activity and expression of IL- 12 were inhibited by sinomenine at concentration ranging from 0.05 - 0.3 mmol/L. Positive correlation was found between the activity of nuclear factor kappa B and the expression of IL - 12. Conclusion Sinomenine inhibit antigen presenting function of dendritic cells by decreasing its nuclear factor kappa B activity.
