血小板生长因子(thrombopoietin,TPO)是一种正常生理状态下的造血生长因子,它刺激造血祖细胞向巨核细胞的分化及血小板的生成和释放。该实验从6月龄人胚肝脏中分离RNA,经逆转录PCR等步骤克隆出了TPOcDNA,并进行了序列分析。将TPOcDNA亚克隆至哺乳动物细胞瞬时表达载体pCMV4,形成了重组表达质粒pCMV4/TPO。以该重组质粒转染COS7细胞。
Thrombopoietin (TPO) is a normal physiological hematopoietic growth factor which is able to induce the hematopoietic progenitor cells to undergo proliferation and differentiation into megakaryocytes and to stimulate the production and release of platelets. The authors report here the cloning of rhTPO from 6 month fetal liver cell mRNA by RTPCR. The cloned rhTPO cDNA has the same sequence as that reported by Bartley. A recombinant expressing plasmid pCMV4/TPO was also constructed by cloning the rhTPO cDNA in an eukaryotic expressing vector pCMV4. When the recombinant plasmid was used to transfect the COS7 cells, temporary expression of rhTPO was detected.
