摘要
本文报道了抗HSVMcAb与抗病毒药物阿糖胞苷偶联物的制备及鉴定。首先用高碘酸钠氧化葡聚糖为多醛基葡聚糖,使之作为中间载体将阿箱胞苷与McAh偶联,经鉴定偶联物中McAb含量为10mg/ml,阿糖胞苷含量为1.05mg/ml,阿糖胞苷与McAb的充分子比为1:60,偶联物的ELISA效价为1:256与偶联前效价相同。应用偶联物和阿糖胞苷分别结膜下注射治疗免眼HSK模型共36只眼,表明偶联物的疗效优于阿糖胞苷。
in this paper, we report that cytosine arabinoside (CA) was coupled to anti-HSVmonoclonal antibody and the conjugates were characterized. Firstly, dextran was oxidized to polyaldehyde dextran by sodium periodate, and then, CA was conjugated to McAb by poly aldehydedextran. The concentration of the McAb and CA in the conjugates were 10 mg/ml, and 1. 05mg/ml, respectively. the molar ratio of CA to McAb was 1: 6. The conjugate showed a higherimmunological activity, and its titer by ELISA was 1: 256. The conjugates and cytosine arabinoside were administrated to eyes of rabbits with HSK by subconjugatival injection respectively.The therapeutic effect of the conjugates was better than that of cytosine arabinoside alone.
