快速检测布鲁氏菌抗体的滴金免疫测定法的建立及应用 被引量:15

摘要 目的:建立一种快速诊断布鲁氏菌感染的方法。方法:将布鲁氏菌可溶性菌体抗原结合于硝酸纤维膜上,以胶体金颗粒结合的羊抗人IgG作为标记抗体,建立滴金免疫测定法,检测布鲁氏菌感染者血清抗体,通过布鲁氏菌抗原抗体渗滤在硝酸纤维膜上反应,2 m in后肉眼观察结果。并与试管凝集试验比较。结果:滴金免疫测定法的敏感性为95.35%(41/43),特异性为98.99%(98/99),其他病患者血清反应均为阴性,Youden′s指数为0.9434。滴金免疫测定法与试管凝集试验的符合率为97.89%(139/142),两法无显著性差异(P>0.05)。结论:滴金免疫测定法可用于布鲁氏菌病的临床检测和流行病学筛查,具有推广应用价值。 Objective: To establish a method to rapidly detect the antibodies in sera of patients with Brucella infection. Methods: Antigen of Brucella was coated on the nitrocellulose membrane, and combined with colloid gold labeled goat anti human IgG, thus the dot- immunogold filtration assay(DIGFA) for rapid Brucella antibodies detection was established, This method was applied to delection Brucella antibodies of patients and the outcomes could be observed after 2 minutes by naked eyes. 142 clinical serum samples, including 43 patients with Brucella infection, 66 healthy people, 33 patients with other diseases were examined by this method. The results of this method were compared with the results of SAT. Results:The sensitivity and specificity of dot - immunogoht filtration assay were 95.33% (41/43) and 98.99% (98/99), respectively. Youden's index were 0. 9434. The coincidence rate between dot - immunogold filtration assay and SAT was 97.89% ( 139/142), and the difference in two methods ( P 〉 0. 05 ) was not significant. Conclusion: Dot - immunogold filtration assay showed promising result for the screening and diagnosis of Brucellosis.
