葛根素对脑缺血再灌注家兔脑组织及内皮细胞的保护 被引量:16

摘要 目的:观察异黄酮类化合物葛根素对家兔脑缺血再灌注损伤血浆丙二醛、超氧化物歧化酶、内皮素及降钙素基因相关肽水平的影响。方法:①实验于2004-03/2004-09在华中科技大学同济医学院附属同济医院麻醉学实验室完成。选用健康家兔24只。随机将兔分为3组:假手术组、缺血再灌注组和葛根素处理组,每组8只。②3组均按“六血管”模式分离颈内动脉、外动脉和椎动脉。缺血再灌注组和葛根素处理组脑缺血30min再灌注4h。葛根素处理组阻断动脉前静注葛根素5mg/kg,注毕阻断上述6条动脉,继之以微量泵150μg/(kg·min)输注至实验结束,缺血再灌注组不用药。③用比色法测定缺血前15min和再灌注4h2个时点血浆丙二醛、超氧化物歧化酶,用放免法测定血浆内皮素、降钙素基因相关肽水平。④组间计量资料差异比较采用方差分析。结果:兔24只均进入结果分析。①再灌注4h血浆丙二醛水平:缺血再灌注组明显高于其他2组(P<0.01),缺血再灌注组和葛根素处理组明显高于缺血前15min(P<0.01)。②再灌注4h血浆超氧化物歧化酶水平:缺血再灌注组明显低于其他2组(P<0.01),缺血再灌注组和葛根素处理组明显低于缺血前15min(P<0.01)。③再灌注4h血浆内皮素水平:缺血再灌注组明显高于其他2组(P<0.01),缺血再灌注组明显高于缺血前15min(P<0.05)。④再灌注4h血浆降钙素基因相关肽水平:缺血再灌注组明显低于其他2组(P<0.01),缺血再灌注组明显低于缺血前15min(P<0.05)。结论:葛根素能降低缺血再灌注期家兔血浆丙二醛、内皮素的含量,提高超氧化物歧化酶的活力,促进降钙素基因相关肽的释放,减轻脑缺血再灌注损伤。 AIM: To observe the effect of puerarin on levels of plasma malondialdehyde (MDA), superoxide dismutase (SOD), endothelin (ET) and calcitonin gene-related peptide (CGRP) in rabbits with cerebral ischemia-reperfusion injury. METHODS: (1)The experiment was conducted in the Laboratory of Department of Anesthesiology, Affiliated Tongji Hospital, Tongji Medical College, Huazhong University of Science and Technology between March and September 2004. Twenty-four healthy rabbits were randomly divided into sham operation group, (group A, n=8), ischemia-repeffusion group (group B, n=8) and puerarin group (group P, n=8). (2)Arteria carotis interha, bilateral external, internal carotid arteries and vertebral arteries in three groups were clipped according to the "six blood vessels" model. Thirty minutes after cerebral ischemia, rabbits in the group B and group P were treated with reperfusion for 4 hours. Rabbits in group P received intravenous injection of puerarin (5 mg/kg) before the artery was occluded, then intravenously injected by micro pump 150 μg/(kg·min) till the end of experiment following the occlusion of above six arteries. Rabbits in the group B received no administration.(3)The levels of plasma MDA and SOD were determined chromatometry, the levels of ET and CGRP were determined with radioimmunological assay (RIA) 15 minutes before occlusion and 4 hours after reperfusion. (4)Differences of measurement data among groups were compared with analysis of variance. RESULTS: A total of 24 rabbits were involved in the analysis of results. (1)Level of plasma MDA 4 hours after reperfusion: It was significantly higher in group B than that in the other two groups (P〈0.01), and it was significantly higher in group B and group P than 15 minutes before ischemia (P〈0.01). (2)Level of SOD 4 hours after reperfusion: It was significantly lower in group B than that in the other two groups (P〈0.01), and it was obviously lower in group B and group P than 15 minutes before ischemia (P〈0.01). (3)Level of plasma ET 4 hours after reperfusion: It was significantly higher in group B than that in the other two groups (P〈0.01), and it was obviously higher in group B than 15 minutes before ischemia (P〈0.05). (4)Level of CGRP 4 hours after reperfusion: It was significantly lower in group B than that in the other two groups (P〈0.01), and it was obviously lower in group B than 15 minutes before ischemia (P〈0.05). CONCLUSION: Puerarin can reduce the content of plasma MDA and ET, increase the activity of SOD, promote the release of CGRP as well as lessen the cerebral ischemic reperfusion injury.
