摘要 目的 探讨泥蚶提取物——海生素抗肿瘤的作用。方法 采用四氮唑盐衍生物(XTT)比色法观察体外培养的肿瘤细胞株Ketr-3和A549,细胞增殖情况,测定细胞蛋白含量和周期分布。计算小鼠移植性S180瘤和艾氏腹水瘤(EAc)的抑瘤率和小鼠生存时间及免疫功能。结果 海生素对Kerr-3和A549细胞具有明显的抑制作用,分别阻滞在细胞G0~G1期和G2~M期。对小鼠S180瘤的抑瘤率达到19.34%~55.19%,明显延长EAC小鼠的生存时间,结论 海生素对不同肿瘤细胞的抑制作用机制可能不同,对移植性肿瘤抑制作用与其免疫功能提高有关。 Objective: To investigate the antitumor effect of haishengsu, an extractive from arca granosa linnaeus, in vitro and in vivo. Methods : The proliferation of the cultured tumor cell lines of Kerr-3 and A549 were evaluated by XTT method. The protein concentration of the cells and the cell cycle distribution were calculated. Transplanted tumor models of S180 and ehrlich ascites carcinoma (EAC) in mice were used to evaluate the antitumor effect of the extractive. Results: The inhibition rates on Ketr-3 and A549 cells growth were directly proportional to haishengsu concentrations. It caused a significant G0~G1, G2~M phase arrest in Ketr-3 and A549 cells respectively. Haishengsu injection could inhibit the Sis0 by 19.34%~55.19% and significantly prolong the life span of EAC-bearing mice. Conclusion: Haishengsu could inhibit tumor and the mechanisms might be both inhibiting the growth of tumor cells directly and strengthening immune system.
