先用0.5%醋酸煮沸样品15min,抽滤,滤渣用0.5%醋酸洗涤,再将滤渣置于消化管中,加入0.1gHgO、1.0gK2SO4和5mL浓硫酸消化。然后采用连续流动分析法测定烟叶样品中的蛋白质含量。方法的RSD为1.1%,回收率在98%~102%之间。并采用该法测定了部分烟叶的蛋白质含量。
The tobacco sample was extracted with 0.5 % acetic acid boiled for fifteen minutes, filtrated, and washed with 0.5 % acetic acid, during which the water soluble nitrogen was removed. Then, the sample residue was digested in the presence of 0.1g HgO, 1.0g K2SO4 and 5mL concentrated sulphuric acid. The protein in resultant digestion solution was determined with an autoanalyzer. The results showed that the relative standard deviation of the method was 1.1%, and the recoveries were from 98% to 102%. The protein in some tobacco leaves were also determined.
