摘要
目的探讨其聚乙烯亚胺(polyethylenimine,PEI)在转导浓度下对肿瘤细胞的毒性作用。方法以PEI为载体将增强型绿色荧光蛋白(enhanced green fluorescence protein,EGFP)报告基因转入A549肺癌细胞,应用台盼蓝染色法和四甲基偶氨唑盐(MTT)法研究不同浓度PEI下A549肺癌细胞的存活率。结果粒径86.9 nm PEI 4μg介导2μg的DNA对肺癌A549细胞的转染率最高(29%),与其余转染梯度相比有统计学差异(P<0.05);台盼蓝染色法检测和MTT法检测不同浓度PEI纳米凝胶下肺癌A549细胞的存活率与对照组间的差异无统计学意义,不同浓度间亦无差异。结论PEI是有效安全的基因转导载体。
Objective To investigate the cytoxicity effect of polyethylenimine on tumor cell under different transduction concentration. Methods A plasmid vector containing the EGFP gene was cultured with PEI of different concentration to observe the survival rate of A549 lung cancer cell. Results Cell transfection rate of A549 lung cancer cell was the highest with 4pg PEI to mediate 2pg DNA evaluated by Trypan blue staining and MTT assay; the survival rate of of A549 lung cancer cell was not significantly different between the various concentration of PEI and control group. Conclusion The PEI is a useful and safe vector for the gene transduction.
