摘要
目的研究毫米波和ADM分别单独应用以及联合应用对K562细胞株培养体系中的氧化自由基及抗氧化酶系的调控和影响,来探讨毫米波及ADM对白血病细胞的作用机制,为研究白血病细胞体外净化方法提供理论依据。方法将毫米波和阿霉素分别单独以及联合作用于K562细胞株,测定细胞培养体系中超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)的活性,测定脂质过氧化产物丙二醛(MDA)的含量。结果单用ADM或毫米波辐射均对K562细胞内MDA的含量有显著的增高(P<0.01);对K562细胞内SOD、GSH-Px两种重要的抗氧化酶系的活性有显著的抑制作用(P<0.01)。毫米波与ADM联合应用能产生协同作用(P<0.01)。结论毫米波和阿霉素联合作用于人白血病K562细胞能产生协同治疗效应,值得进一步研究。
AIM: To study the effect of combination millimeter wave with adriamycin on regulating the antioxidase system and oxyradical in the cultured cell line system in vitro. METHODS: Leukemic cell line K562 was treated by millimeter wave alone and in combination with adriamycin. Then Intracellular malondialdehyde (MDA) was analyzed with thiobarbituric acid method, superoxide dismutase (SOD) was analyzed with pyrogallic acid method, and glutathione peroxidase (GSH-Px) was analyzed with improved Hafeman method. RESULTS: The combination of millimeter wave and adriamyein had a synergistic effect on inhibiting superoxide dismutase and malondialdehyde of leukemie eell line K562. The activity of SOD and GSH-Px was deereased and the eontent of MDA was improved by the combination. CONCLUSION: The combination of millimeter wave and adriamyein shows a synergistic effect on puring leukemic cells in vitro.
