摘要
目的:探讨半胱氨酶-3(Caspase-3)在二乙酰二脱水卫矛醇(d iacetyld ianhydrogalactitol,DADAG)诱导小鼠白血病L1210凋亡过程中的作用。方法:MTT法观察DADAG对L1210细胞的生长抑制作用;透射电镜检测细胞凋亡;Caspase-3试剂盒测定细胞内Caspase-3的酶活性。结果:DADAG对L1210细胞有较强的体外增殖抑制作用,其IC50值为24.6 mg/L;DADAG可诱导L1210细胞发生凋亡及时间依赖性地升高Caspase-3酶活性。结论:Caspase-3在DADAG诱导的细胞凋亡中可能起重要作用。
Objective: To investigate the role of Caspase-3 in DADAG-indueed apoptosis of mouse L1210 leukemia. Methods: The growth inhibiton was analysed by MTT assay. DADAG-induced apoptosis in L1210 ceils was identified by electron microscopy; Caspase-3 activity was measured by ApoAlert CPP32 colorimetrie assay kit. Results: DADAG showed potent antiproliferation on L1210 cells with the IC50 of 24.6 mg/L in vitro. Meanwhile apoptosis was induced in targeted cells. DADAG time-dependently increased the activity of Caspase-3. Conclusion: Caspase-3 might play an important role in DADAG-induced apoptosis of L1210 ceils.
