TGF-β1对老年人牙周膜细胞分化功能的影响 被引量:1

摘要 目的:探讨转化生长因子-β1(TGF-β1)对老年人牙周膜细胞碱性磷酸酶活性和骨钙素分泌的影响。方法:将不同浓度的TGF-β1(0.5μg/L,1μg/L,2μg/L)与体外培养的老年人牙周膜细胞一同培养后,采用酶动力学方法和放射免疫法检测碱性磷酸酶活性和骨钙素分泌水平,并与青年人牙周膜细胞进行比较。结果:①在老年组,TGF-β1各浓度组细胞裂解液和培养液中碱性磷酸酶活性均明显高于对照组(P<0.05,P<0.01)。老年组和青年组差值比较,除TGF-β1(0.5μg/L)组细胞培养液外,无论是细胞裂解液还是培养液中碱性磷酸酶活性青年组均明显高于老年组(P<0.05,P<0.01)。②在老年组,只有7 d时的TGF-β1(0.5μg/L)组骨钙素分泌高于对照组(P<0.05);在青年组,7 d时各组和21 d时TGF-β1(1μg/L)组骨钙素分泌均明显高于对照组(P<0.05,P<0.01)。老年组和青年组差值比较,7 d时TGF-β1(1μg/L)、(2μg/L)组和21 d时的TGF-β1(0.5μg/L)、(1μg/L)组骨钙素分泌青年组明显高于老年组。结论:合适浓度的TGF-β1不仅可以促进青年人而且可以促进老年人牙周膜细胞向成骨样细胞转化,增强其成骨能力;其对青年人牙周膜细胞的作用强于老年人。 AIM:To investigate effects of TGF- β1 on alkaline phosphatase activity and the osteocalcin secretion of periodontal ligament cells(PDLCs) in elderly people. METHODS :The PDLCs of elderly and young people were isolated and cultured. The effects of TGF - β1 (0.5 μg/L, 1 μg/L,2 μg/L) on alkaline phosphatase activity of PDLCs in elderly people were measured with alkaline phospbatase activity assay kit; the osteocalcin secretion of PDLCs in elderly people was determined by radioimmunoassay with osteocalcin immunoradiometric kit. The results of elderly people were compared with those of young people. RESULTS :①Comparing with the control group, alkaline phosphatase activities of PDLCs treated with different concentrations of TGF - β1 in elderly people groups were significantly increased( P 〈0.05 ,P 〈0.01 ). not only in lysis buffer but also in culture media. Comparing the differences of alkaline phosphatase activities between the groups treated with TGF- β1 and control groups, the alkaline phosphatase activities both in lysis buffer and in culture media in young people were significantly higher than those in elderly people ( P 〈 0.05, P 〈 0.01 ) except in cell culture medium treated with TGF - β1 (0.5 μg/L). ②Comparing with the control group, the osteocalcin secretions of PDLCs treated with TGF - β1 (0.5 μg/L) in elderly people on day 7 and the osteocalcin secretions of PDLCs treated with TGF - β1 ( 1 μg/L) in young people on clay 7 and day 21 were obviously increased(P 〈 0. 05 ). Comparing the differences of the osteocalcin secretions between the groups treated with TGF - β1 and control groups, the osteoealcin secretions of PDLCs treated with TGF - β1 ( 1 μg/L,2 μg/L) on day 7 and with TGF - β1 (0.5 μg/L, 1 μg/L) in young people were significantly higher than those in elderly people. CONCLUSION :These findings suggested that the TGF- β1 can promote PDLCs to differentiate in elderly and young people. This effect is more significant in young people than in elderly ones.
