摘要
目的探讨细胞凋亡与STZ糖尿病大鼠视网膜微血管病变之间的关系。方法建立糖尿病大鼠动物模型,随机分为糖尿病3月组(DM3)及6月组(DM6),制备视网膜消化铺片,采用核苷酸末端转移酶介导DUTP缺口翻译法(TUNEL)检测视网膜毛细血管细胞凋亡变化,PAS染色及图像分析,动态观测视网膜微血管形态的改变。同时设立正常对照组。结果3月时糖尿病大鼠视网膜微血管周细胞发生细胞凋亡,周细胞数目减少,对照组及DM3组周细胞数目分别为(6.71±0.45)个及(6.17±0.45)个,2组比较差异具有显著性(P<0.01);6月时细胞凋亡呈增加趋势,DM6组及对照组周细胞数分别为(5·66±0·60)个及(6.52±0.40)个(P<0.01);与DM3组相比DM6组周细胞数目明显减少(P<0.01)。内皮细胞各组间差异均无显著性(P>0.05)。无细胞毛细血管数目增多。结论糖尿病大鼠视网膜周细胞凋亡呈增加趋势,并与微血管病变有关。
Objective To explore the relationship between apoptosis and retinal microvascular pathological changes in STZ diabetic rats. Methods Diabetic animal models were induced by STZ on SD rats. The rats were randomly divided into 3 months (DM3) and 6 months (DM6) group. Retinal capillary trypsin digestion were prepared to observe the microvascular shape. TUNEI, method was used for detection of apoptosis. Control group was established simultaneously. Results At the 3rd month, retinal capillary perlcyte in diabetic rats was found apoptosis and the pericyte number was decreased sharply. The pericyte number of control group and DM3 group were 6.71±0.45 and 6.17±0.45 respectively, and there was obvious difference between two groups(P〈0.01 ). At the 6th month, apoptosis was increased significantly. The pericyte number of control group and DM6 group were 5.66±0.60 and 6.52± 0.40 respectively, and there was obvious difference (P〈0.01 ). Compared with DM3, the perlcyte number of DM6 group decreased sharply ( P〈0.01 ). There was no change of endothelial cells in each group( P〉0.05 ). Acellular capillaries were formed and increased. Conclusion Apoptosis shows an increase trend in diabetic rats and be relevant to the microvascular pathologica changes.
