摘要
目的:寻找诊断华支睾吸虫病的特异性抗原。方法:应用SDS-PAGE和Western blot对华支睾吸虫成虫可溶性抗原中的蛋白组分进行分析,试验血清包括旋毛虫感染的大鼠和小鼠血清,正常大鼠和小鼠血清,旋毛虫病、华支睾吸虫病、并殖吸虫病、日本血吸虫病及囊虫病患者血清,斯氏狸殖吸虫感染的大鼠血清和正常人血清。结果:华支睾吸虫成虫可溶性抗原蛋白经SDS-PAGE后显示17条蛋白带,其中相对分子质量为65000、54000、31000、25000、13000的为主带。Western blot结果显示,相对分子质量为108000、94000、71000、65000、56000、53000、43000的蛋白带可被华支睾吸虫病患者血清识别,108000、71000、65000、63000、53000的蛋白带可被并殖吸虫病患者血清识别,108000、94000、71000可被日本血吸虫病患者血清、旋毛虫感染的大鼠血清及旋毛虫病患者血清识别;56000的蛋白组分只能被华支睾吸虫病患者血清识别,而不与其他试验血清发生交叉反应。结论:华支睾吸虫成虫可溶性抗原中相对分子质量为56000的蛋白组分为华支睾吸虫成虫的特异性抗原。
Aim: To find out the specific antigens for immunodiagnosis of clonorchiasis. Methods: The soluble antigens of Clonorchis sinensis(C.sinensis)adult worms wrer analyzed by SDS-PACE and Western blot. Sera for this study included sera from patients with clonorchiasis,paragonimiasis,schistosomiasis,trichinellosis,sera from rats and mice infected with T.spicalis,and sera from normal rats, mice and persons. Results:SDS-PAGE revealed that the soluble antigens of C.sinensis adult worms had 17 protein bands.Westerm blot results showed that the protein bands with MW 108 000, 71000,65000,63000,53000 were crose-reacted with sera from patients with paragonimiasis. All of the protein bands with MW 108000,94000,71000 were cross-reacted with sera from patients with schistosomiasis, rats infected ith T.spiralis and patients with trichinellosis. The pratein component with MW 56000 was only reacted with sera from patients with clonorchiasis.Conclusion:The protein component with MW 56000 is the specific antigen for C.sinensis adult worm.
