摘要
目的建立大鼠非酒精性脂肪性肝炎(NASH)模型。方法将♂SD大鼠,随机分为正常组(n=15)和模型组(n=15)。两组动物均给以普通饲料,模型组每天ig高脂、高糖、高蛋白乳剂,剂量递增。实验持续6周后,检测血清ALT、AST、TC、TG、HDL-C、LDL-C及肝脏TC、TG,并行病理组织学检查。结果模型组大鼠血清ALT、AST、TC、HDLC、LDL-C及肝脏TC、TG均显著增高;病理组织学检查提示模型组大鼠肝细胞呈现弥漫性脂肪变性伴肝小叶炎症坏死。结论利用改良乳剂灌胃的方法,成功复制出大鼠NASH模型。
Objective To establish a rat model of nonalcoholic steatohepatitis. Methods The model rats were fed with high fat, high sucrose and high protein emulsion via gavage (from 2. 5 - 10 ml/kg, once a day) and standard rodent chow adlibitum for 6 weeks. Serum levels of ALT, AST, TC, TG, HDL, LDL, hepatic TC and TG content and histopathologic changes of liver were evaluated. Results The serum levels of ALT, AST, HDL, LDL, TC, TG, hepatic TC and TG content in model rats were significantly higher than those in normal rats. The histopatholog- ical analysis suggested that the model rats developed panlobular steatosis and inflammatory cell infiltration. Conclusion A NASH model is successfully induced in rats by feeding with high fat, high sucrose and high protein emulsion via gavage .
