摘要
目的系统分析ndrg2在胎前脑组织中表达分布的时空特点,以期明确ndrg2在胎前脑脑功能区的组织分布、细胞定位及表达趋势,为进一步研究其功能提供实验依据。方法收集引产4 ̄7月胎脑组织标本,分别进行组织石蜡切片和总RNA提取,应用免疫组化方法和RT-PCR技术检测ndrg2在蛋白质及mRNA表达水平变化规律。结果免疫组化结果表明,在上述胎前脑脑区神经元细胞胞浆中均有ndrg2蛋白不同程度的表达。RT-PCR结果显示,ndrg2mRNA在正常胎脑各脑区均有表达;随脑组织发育成熟,ndrg2表达水平增高,4月胎脑中海马ndrg2表达水平较高,而到7月额叶、顶叶则表达增强。以上结果表明,ndrg2基因在不同脑功能区广泛表达,但不同时期、不同胎脑功能区域表达有明显区别。结论在脑发育的过程中,ndrg2表达呈一定时空特点,ndrg2可能对神经细胞发育成熟有一定影响。
Objective To explore the temporal and spatial expression features of ndrg2 during human embryonic forebrain development and to investigate the distribution, cellular localization and expression tendency of ndrg2 in functional domain of embryonic forebrain, so as to provide some clues for further study on the function of ndrg2. Methods 4^th-7^th month human embryonic brain tissues of induced abortion were collected for extracting total RNA or preparing paraffin section of brain tissues. Immunohistochemistry and reverse transcriptasepolymerase chain reaction (RT-PCR) were used to examine the expression of ndrg2 in both mRNA and protein. Results Immunohistochemistry showed that ndrg2 protein was extensively expressed in neuronal cytoplasm of the 4^th-7^th month human embryonic forebrain tissues. RT-PCR indicated that ndrg2 mRNA was found to be extensively expressed in these normal 4^th-7^th month human embryonic forebrain tissues. With the development of human embryonic brain tissues, ndrg2 expression in functional domain of embryonic forebrain increased and reached a high level in hippocampus of 4^th month embryonic brain, and then increased in frontal lobe and parietal lobe of 7^th month embryonic brain. These results indicated that ndrg2 was extensively expressed in functional domain of these 4^th-7^th month normal human embryonic forebrain, but varied in different areas. Conclusion During the development of human embryonic forebrain, ndrg2 has shown the character of the temporal and spatial expression. With the development of human embryonic brain, ndrg2 expression in functional domain of embryonic forebrain increased. Ndrg2 might play an important role in the development and the maturity of human brain.
