巨噬细胞移动抑制因子在幽门螺杆菌相关胃粘膜炎症中的表达 被引量:4

摘要 目的检测MIF在幽门螺杆菌相关胃粘膜炎症中的表达,并在体外分析幽门螺杆菌感染对单核细胞MIF表达的影响。方法连续收集胃镜检查的标本,胃窦与胃体同时进行病理组织学检查,根据悉尼系统作胃粘膜的组织学分型。MIF/T细胞(CD45RO)和MIF/巨噬细胞(KP1)进行双重免疫组化染色,MIF mRNA进行原位杂交检测。在体外幽门螺杆菌ATCC26695与THP-1单核细胞共培养后,分别用ELISA与RT-PCR测定THP-1细胞MIF的表达。结果在62例慢性胃炎患者中,42例幽门螺杆菌阳性患者胃窦与胃体粘膜内T细胞总数、MIF阳性T细胞数、巨噬细胞总数、MIF阳性巨噬细胞数和MIF mRNA阳性细胞数都显著高于20例幽门螺杆菌阴性患者。而且在42例幽门螺杆菌相关胃粘膜炎症中MIF的表达随炎症程度的增加而增加。在体外幽门螺杆菌与THP-1细胞共培养后,幽门螺杆菌促进了THP-1细胞MIF蛋白质与mRNA表达增加。结论MIF在幽门螺杆菌感染相关胃粘膜炎症细胞中表达增加,可能在幽门螺杆菌感染相关胃粘膜炎症的形成中起着重要的作用。 This study aimed to determine macrophage migrotion inhibitory factor MIF expression level in H. pylori induced gastritis, and the effect of H, pylori promoting MIF expression of monocytes in vitro was analysed. Gastric biopsies were collected by using endoscopies, Histopathological examination was carried out to check gastric antrum and body. The update Sydney System was employed to assess the severity and activity of gastric inflammation. Double immunoassaying for MIF/T-cells (CD45RO) and MIF/rnacrophage (KP1), as well as in situ hybridization for the expression of MIF mRNA were carried out. THP-1, a monoeyte cell line, was co-incubated with H. pylori strains (ATCC26695) and suhsequendy examined for the expression of MIF protein and mRNA by enzyme linked immunosorbent assay and retrospective transcription-polymerase chain reaction, respectively. The results indicated that among 62 patients with chronic gastritis, significant increase in total T-cells, MIF+T-cells, total macrophages, MIF+ macrophages and MIF mRNA+cells was observed in 42 H. pylori positive patients with compared to 20 H. py/or/ negative patients. Moreover, the increase of the MIF mRNA + cells was highly correlated with the severity of the disease . In vitro results showed that the expression of MIF protein and mRNA in monocytes was significantly increased after incubation with H. pylori strains. In Conclusiou H. pylori infection stimulates the expression of MIF in the gastric inflammatory cells, which may play a significance role in H. pylori-induced gastric inflammation.
