摘要
目的 研究重组血管基膜衍生多功能肽(vascular basementme mbranederived muldfunction pepude,VBMDMP)的抗人结肠癌作用。方法 体外培养人结肠癌细胞系(SW480)细胞和中国仓鼠卵巢细胞系(CHO-K1)细胞。采用MTT比色法检测重组VBMDMP对SW480细胞选择性抑制增殖作用;胎盼蓝染色细胞计数法检测重组VBMDMP对SW480细胞和CHO-K1细胞生长曲线的影响;人结肠癌(SW480)细胞裸鼠异种移植瘤模型治疗实验观测重组VBMDMP体内抗人结肠癌作用。结果 重组血管基膜衍生多功能肽具有选择性抑制体外培养人结肠癌细胞系SW480细胞增殖和生长作用,呈剂量依赖性。而对CHO-K1细胞的增殖活性影响小。重组血管基膜衍生多功能肽对SW480细胞裸鼠异种移植瘤生长具有显著抑制作用.呈剂量依赖性。1、5、10mg/kg重组VBMDMP的肿瘤生长抑制率分别为:63%,79%和83%;瘤重抑制率分别为:62%、66%和75%。10mg/4kg重组VBMDMP的肿瘤生长抑制率和瘤重抑制率均接近100mg/kg环磷酰胺(CTX),高于20ms/kg 血管生成抑制剂烟曲霉素衍生物TNP-470。结论 重组血管基膜衍生多功能肽对人结肠癌细胞增殖和生长具有显著抑制作用。
Objective To investigate the inhibitory effect of recombinant vascular basement membrane derived maltifunction pepfide on human colon cancer. Methods Human colon cancer (SW480) cells and Chinese hamster ovary (CHO - K1 ) cells were cultivated in vitro. Recombinant VBMDMP selectively inhibited the proliferation of SW480 cells detected by MTT assay. Trypan blue exclusion method was used to test the influence of growth curve in SW480 cells by recombinant VBMDMP. Therapeutic trial of human colon cancer xenograft in nude mice was used to investigate the anti - tumor activity of recombinant VBMDMP in vivo. Results Recombinant VBMDMP can selectively inhibit the proliferation and growth of SW480 cells in a dose - dependent manner with little effect on CHO - K1 cells. Recombinant VBMDMP possessed significant inhibitory growth effect of human colon cancer xenograft in nude mice in a dose - dependent manner. The inhibitory rate of xenograft growth was 63%, 79% and 83% treated with recombinant VBMDMP at 1,5,10 mg/kg respectively. The inhibitory rate of xenograft weight was 62 % ,66% and 75% treated with recombinant VBMDMP at 1,5,10 mg/kg respectively. The inhition effect of xenograft growth and xenograft weight with recombinant VBMDMP at 10 mg/kg is close to cyclophesphamide(CTX) at 100 mg/kg and exceed angiogenic inhibitor TNP- 470, an analogue of fumagillol at 20 mg/kg. Conclusion Recombinant vascular basement membrane derived multifunction peptide possess significant inhibitory effect on proliferation and growth of human colon cancer cells.
