摘要
从牡丹皮中提纯的丹皮酚结晶(0.6 mg.mL-1)与烟草花叶病毒(Tobacco m osaic virus,TMV)混合10 m in后对TMV枯斑抑制率达80%以上,说明丹皮酚对叶圆片中的病毒复制具有抑制作用;电镜下观察丹皮酚与TMV混合对粒体形态没有影响;丹皮酚与TMV混合后透析病毒可恢复侵染力,表明丹皮酚对病毒没有直接毒害作用;用丹皮酚喷施叶面不影响病毒侵染;低含量丹皮酚可阻碍TMV-CP的体外聚合,对TMV-RNA无体外抑制侵染作用.据此推测,丹皮酚对TMV的抑制作用可能主要是通过其与TMV-CP结合而影响病毒粒体对叶片表面侵染位点的识别,或影响植株体内TMV-CP亚基的体内聚合.
The infectivity of Tobacco mosaic virus(TMV) in Nicotiana glutinosa was strongly inhibited (more than 80%) by 0.6 mg·mL^-1 paeonol that was extracted from Paconia suffruticoss after mixing paeonol with TMV for 10 min; the paeonol had inhibiting effect on the replication of TMV in infected leaf discs; there was no virion morphological change observed under electron microscope after TMV mixing with paeonol; the lost infectivity of TMV was recovered by dialysising the virus-paeonol suspension in vitro, which showed that the paeonol had not direct viricidal effect; the paeonol with low content inhibited the polymerization of TMV coat protein in vitro; the paeonol did not decrese infectivity when it was applied before TMV inoculation, or inoculated with TMV-RNA in vitro. It suggested that the paeonol inhibit TMV by binding with TMV coat protein and interfering with the recognition of a host receptor essential for infection or by interfering with TMV-CP polymerization in plants.
