摘要
目的:观察灯盏乙素对大鼠局灶性脑缺血再灌注损伤的保护作用。方法:采用大脑中动脉栓线阻断法制备局灶性脑缺血再灌注损伤模型,用TTC染色检测灯盏乙素对再灌注损伤大鼠脑梗死体积的影响,用流式细胞术检测细胞凋亡百分率,用免疫组织化学和RT-PCR技术检测诱导细胞凋亡Cas-pase-3mRNA及其蛋白的表达。结果:灯盏乙素应用于缺血再灌注损伤大鼠可明显缩小脑梗死体积,降低脑细胞凋亡百分率,抑制脑组织Caspase-3mRNA及其蛋白的表达。结论:灯盏乙素对大鼠局灶性脑缺血再灌注损伤具有保护作用,其作用机制可能与抑制脑缺血后神经细胞凋亡有关,与抑制诱导凋亡Cas-pase-3mRNA及其蛋白的表达有关。
Objective:To investigate the protective effect of scutellarin on focal brain ischemiareperfusion injury in rats. Methods:The models of focal brain ischemia-reperfusion injury were established by middle cerebral artery occlusion in Wistar rats. The infarct volume of brain ischemlc damage was estimated by Trc staining. The apoptosis was determined by flow cytometry (FCM). Caspase-3 protein expression was detected by immunohistochemistry. The expression of the Caspase-3 mRNA was deteced by reverse transcrip- tion polymerase chain reaction (RT-PCR). Results: The infarct volume,the apoptosis,the expression oF Caspase-3 and Caspase-3mRNA in brain of focal cerebral ischemia-reperfusion in rats were decreased by scutellarin. Conclusion: Scutellarin can prohibit the brain damage induced by focal ischemia-reperfusion in rats by inhibiting the neuronal apoptosis and the process of the neuronal apoptosis, as well as inhibiting the transcription of Caspase-3mRNA and the expression of the Caspase-3 protein.
