摘要
目的:研究幽门螺杆菌空泡毒素(VacA)编码基因在大肠杆菌中的表达产物对SGC7901胃癌细胞的作用.方法:常规培养pET32a-vacA-E.coliBL21(DE3)工程菌株,IPTG诱导重组蛋白表达,产物纯化后作用于胃癌细胞,倒置显微镜及电镜等检测细胞生长情况.结果:重组蛋白作用于胃癌细胞后,倒置显微镜下观察,细胞生长受抑制,其细胞空泡毒作用减弱甚至消失,电镜下可见10%-20%左右细胞呈现轻度空泡变,仅少数细胞空泡变明显,细胞表面微绒毛消失,少量细胞体积减小,并出现了早期核着边固缩等凋亡改变.结论:重组VacA蛋白对胃癌细胞的生长具有抑制作用,空泡毒作用减弱.
AIM: To explore the effect of the recombinant protein expressed by Helicobacter pylori (H pylori) VacA gene in E.coli on the gastric cancer cell line SGC7901.
METHODS: The engineered bacteria pET32a-VacA-E.coli BL21 (DE3) was cultured and analyzed. The isopropyl-β-D-thiogalactopyranoside (IPTG) was used to induce the expression of the recombinant protein. Then the purified protein was added into the gastric cancer cells, and the growth and morphological changes of the cell were observed by the inverted microscope and electron microscope.
RESULTS: The proliferation of the gastric cancer cells was inhibited after the recombinant protein was added. The effect of VacA was reduced or even failed. About 10%-20% SGC cells were lightly vacuolated, and only a few of them were severely vacuolated. The microvilli on the cell surface disappeared, and the volume of some cells was decreased. Apoptosis was induced by the recombinant VacA in some SGC cells.
CONCLUSION: The recombinant protein can inhibit the proliferation of gastric cancer cell, and the vacuolated effect becomes weakened and even fails.
