摘要
目的观察左旋卡尼汀对大鼠离体心缺血再灌注损伤的保护作用。方法24只大鼠随机分成2组,对照组:缺血30min,复灌120min;实验组:缺血30min,10mmol/L左旋卡尼汀复灌。记录冠脉流量、左心室发展压(LVDP)、左心室压力时间变化率(±DP/DT);检测冠脉流出液中丙二醛(MDA)、超氧化的歧化酶(SOD)、肌酸磷酸激酶(CPK)、乳酸脱氢酶(LDH)浓度和心肌组织中MDA、SOD含量。结果复灌期间补充左旋卡尼汀可明显改善缺血再灌后的血流动力学变化:±DP/DTmax和LVDP较对照组显著升高,冠脉流量增大;冠脉流出液中MDA、LDH、CPK以及心肌组织中MDA浓度降低,而冠脉流出液和心肌组织中SOD含量均升高。结论左旋卡尼汀具有抗大鼠离体心缺血再灌注损伤的作用。
Objective To investigate the protective effect of L-carnitine on myocardial ischemia-reperfusion injury in rat heart. Methods All harvested isolated hearts were perfused for 20 min on Langendorff apparatus with oxygenized K-H solu tion. The hearts were then exposed to ischemia for 30 mins. Following the ischemia the hearts were reperfused with K-H solution for 120 min to make up the control group. 10 mmol/L of L carnitine was added into the K-H solution for 20 min at the beginning of reperfusion to make up treatment group. ± DP/DT,LVDP and coronary flux were investigated during the entire procedure. The levels of MDA and SOD in tissue, and LDH, CPK, MDA and SOD concentration in the coronary efflux were measured. Results Compared with the control group, the treatment of L-carnitine can lead to a better outcome: + DP/DTmax and LVDP were higher, ventricular fibrillation was reduced, the level of SOD were elevated, while the concentration of MDA was debased, as well as the CPK and LDH. Conclusion L-carnitine has a protective effect against ischemia-reperfusion injury.
