摘要 目的建立测定舍曲林的高效液相色谱法并测定人血浆中舍曲林的含量。方法色谱柱为Agilent Zorbax Extend—C18(4.6mm×250mm,5μm),流动相为乙腈-0.15mol·L^-1 NaH2PO4(pH3.0)=40:60,流量为0.80mL·min^-1,紫外检测波长230nm。结果舍曲林浓度在1.0~150.0ng·mL^-1内,线性关系良好(γ=0.9999);最低检出限为1ng·mL^-1;平均回收率为(100.54±4.01)%,日内、日间精密度均〈5%。结论本方法简便、准确、灵敏.可用于舍曲林浓度测定。 Objective To establish a high performance liquid chromatography method for the determination of sertraline in healthy human plasma. Methods Agilent 1100 series HPLC system was applied with the column Zorbax Extend - C18 ( 4. 6 mm ± 250 mm, 5μm ). The mobile phase was composed of methanol (0.15 mol·L^-1 ) and NaH2PO4 ( pH 3.0 ) with ratio of 40: 60, and the flow rate was 0.80 mL · min^-1. The UV detection wavelength was 230 nm. Results The calibration curve was liner in the range from 1.0 to 150.0μg · L^-1 (γ= 0.9999 ) , with determination limit of 1 μg·L^-1 and the average recovery of [ 100.54 ± 4.01 ) %. The intra - day RSD and inter - day RSD were all less than 5%. Conclusion The method was simple ,rapid , accurate and can be used to determine the sertraline concentration in plasma.
