摘要
目的 观察缺血再灌注模型大鼠脑内N-甲基-D-天门冬氨酸受体1(NMDAR1)mRNA的变化,探讨胡黄连对神经元的保护作用。方法 参照线栓法制作大鼠脑缺血再灌注模型,胡黄连干预,原位杂交方法观察皮质、纹状体NMDAR1 mRNA表达。结果 对照组缺血侧皮质区、纹状体区于缺血再灌注1d时NMDAR1 mRNA表达达高峰,7d后逐渐下降,但各时间点与假手术组比较,均有显著性差异(F=3.6、7.2,q=11.4~16.7,P〈0.01);胡黄连治疗组大鼠脑皮质与纹状体NMDAR1 mRNA基本接近假手术组,明显低于对照组(t=9.43~10.55,P〈0.01)。结论 胡黄连对脑缺血再灌注损伤神经元有一定保护作用,其机制可能与下调脑内NMDAR1 mRNA表达有关。
Objective To observe the expression of N-methyl-D-aspartate receptor 1 (NMDAR1) mRNA following cerebral ischemic reperfusion in rat brain, and to investigate neuroprotective effects of picrorhizae on neurons. Methods The cere bral ischemic-reperfusion models in rat were made by intraluminal-thread method and interfered by picrorhizae. The expression of NMDAR1 mRNA in cortex and striatum were determined with in situ hybridization. Results In the control group, the expression of NMDAR1 mRNA peaked at one day on ischemic cortex and striatum and descended gradually after seven days, Compared with sham-operation group, there was significant difference at each time point (F = 3.6,7.2;q = 11.4- 16.7; P〈0.01 ). In the cortex and striatum of picrorbizae-treated group, the expression of NMDAR1 mRNA positive neurons was close to that of the sham-operation group, but significantly lower than in the control group (t=9.43 10.55, P〈0.01). Conclusion Picrorbizae could protect the neurons from ischemic-reperfusion injury by decreasing the expression of NMDAR1 mRNA in the brain.
