应用实时定量PCR技术检测慢性粒细胞白血病患者bcr/abl^(P190)融合基因转录本 被引量:1

Quantification of bcr/abl^(P190) fusion gene transcripts by real time quantitative PCR in patients with chronic myeloid leukemia

摘要 目的研究bcr/ablP190融合基因转录本在慢性粒细胞白血病(CML)中表达水平及其临床相关性。方法应用实时定量聚合酶链反应(RQ-PCR)检测74例CML患者骨髓单个核细胞中的bcr/ablP190转录本。结果bcr/ablP190转录本阳性54例(77.1%),中位含量2.60(1.08~8.58)×10-3,与性别、年龄、白细胞总数、血红蛋白含量、血小板计数、bcr/ablP210转录本类型和表达水平均不相关,但bcr/ablP190阳性患者的白细胞总数和血小板计数较阴性者明显降低。bcr/ablP190转录本水平在急变期与慢性期患者以及不同类型急变患者中都无差异,而细胞遗传学完全缓解患者的bcr/ablP190水平较慢性期患者、干扰素无效患者和急变期患者则明显下降。结论不同阶段CML患者体内大都存在着不同的bcr/abl剪切型转录本,不同剪切型的存在具有一定的血液学相关性。 Objective To study the expression and its clinical relevance of bcr/abl^p190 fusion gene transcripts in patients with chronic myeloid leukemia (CML). Methods A real time quantitative PCR (RQ-PCR) was performed to measure the bcr/abl^p190 transcripts in the bone marrow mononuclear cells from 74 CML patients in different phases. Results bcr/abl^p190transcripts were detected in 54 (77.1 %) cases. The median bcr/abl^p190 level was 2.60 (1.08-8. 58) 10-a, which was not correlated with the sex, age, leukocyte counts, hemoglobin, platelets, and the types and expression level of bcr/abl^p190 fusion transcripts. The white blood cells and platelets in bcr/abl^p190 positive group were significantly lower than those in bcr/abl^p190 negative group. There were no significant differences in bcr/abl^p190 levels between the patients in blastic crisis and in chronic phase and between the patients with different blastic phenotypes. The bcr/abl^p190 level was decreased in cases obtained complete cytogenetic remission. Conclusion The different types of bcr/abl splicers were present in CML patients in different phases and there may be a certain relevance of splicing and hematological parameters.
