摘要
目的观察低氧诱导因子1α(hypoxiainduciblefactor1α,HIF1α)和促红细胞生长素(erythropoietin,EPO)在前脑缺血大鼠海马中的动态表达规律,为临床开辟治疗脑缺血的新途径时间窗提供依据。方法采用双侧颈总动脉永久性阻断法(2VO)建立大鼠前脑缺血的动物模型,应用免疫组化法检测大鼠海马CA1区HIF1α和EPO的表达水平。结果血管阻断后3h大鼠海马CA1区HIF1α和EPO的表达开始明显升高,1周达到高峰,此后缓慢下降,但仍显著高于假手术组。结论HIF1α/EPO缺氧信号转导系统可能作为保护因子参与了脑缺血后机体的代偿过程。
Objective To observe the expression rule of HIF-1α and EPO in the hippocampus of forebrain ischemia rats, and to provide theoretic foundation for new therapeutics of cerebral ischemia. Methods Rats of experimental group were treated with a permanent bilateral common carotid arteries oeelusion(2-VO) for establishing forebrain isehernia model. The dynamic expression of HIF-1α and EPO in hippoeampal CA1 region was determined by immunohistoehernieal method. Results HIF-1α and EPO positive cells in hippoeampal CA1 region began to increase at 3h after vessel-occlusion, reached its peak level at lw, and then declined. Conclusions HIF-1/EPO anoxie signal transduetion may play a protecting role in the compensatory process after cerebral isehernia.
