摘要
目的:观察清肝活血方药物血清对体外培养肝星状细胞(HSC)增殖及胶原生成的影响。方法:原位分离大鼠肝细胞、HSC3、H-TdR掺入法测定细胞增殖能力,ELISA法测定Ⅰ型胶原含量。结果:乙醛可以直接促进HSC增殖和Ⅰ型胶原的分泌,乙醛损伤肝细胞条件培养液培养下的HSC出现相同的改变,不同剂量的清肝活血方药物血清能产生正向调节作用,有效地抑制HSC增殖和Ⅰ型胶原分泌。结论:清肝活血方既可以通过拮抗乙醛引起的肝脏脂质过氧化损伤,还可以直接阻止乙醛引起的HSC的活化增殖及Ⅰ型胶原的分泌,从而有效预防肝纤维化的发生和发展。
Objective: To observe the effect of Qingan Huoxue decoction serum drug on proliferation and collagen synthesis in hepatic stellate cells. Methods; Hepatocyte and hepatic stellate cells were isolated form Wistar rats'livers. Cell proliferation ability was measured by^3H-TdR method. The enzyme-linked immunoabsorbent assay (ELISA) detected the content of type Ⅰ collagen. Results: Acetaldehyde can directly accelerate hepatic stellate cells proliferation and type Ⅰ collagen secretion. The same alteration of hepatic stellate cells was observed when cultured with the media pretreated with hepatocyte exposed to acetaldehyde. Various dosage of Qingan Huoxue decoction serum drug developed positive regulation of inhibiting hepatic stellate cells proliferation and type Ⅰ collagen secretion. Conclusion: Qingan Huoxue decoction prevented the process of liver fibrogenesis via interacting lipid peroxidation damage liver caused by acetaldehyde and directly preventing hepatic stellate cells proliferation and type Ⅰ collagen secretion.
