富亮氨酸重复序列(Leucine—rich-repeat,LRR)是一高度保守的氨基酸序列,通常由20-29个氨基酸残基组成,11个氨基酸高度保守。晶体结构分析表明LRR是由一个β片层和一个α螺旋通过loop环连接形成的一个非球形的、马蹄彤分子。序列分析显示LRR蛋白超家族至少包含7个不同的亚家族。LRR蛋白的表达定位多种多样,LRR蛋白具有广泛的生物学作用。在这些生物学过程中,LRR结构域主要介导蛋白与蛋白之间的相互作用。
Leucine-rich repeat (LRR) motif is a highly conserved sequence which span a variety of species and throughout species evolution. The LRRs are generally 20 -29 residues long and contain a conserved l 1-residue segment with the consensus sepcence LxxLxLxxN/CxL. The crystal structures analysis of LRR indicated each LRR consist of a β strand( LxxLxLxxN/C xL) and an α helix (xaxx ±α ± ± ± ± α ± ± x±± ) connected by loops and forming conformed a nonglobular, horseshone-shaped molecule. Sequence analyses of LRR proteins revealed there are at least seven distinct subfamilies in the LRR superfamily. The location of expression of the LRR proteins is very diverse and have been found in the se- rum, extracellular matrix, cytoplasm, nucleus, and extracellular and intracellular domains of membrane proteins, and LRR proteins as protein recognition motifs participate in ubiquitous biological processes. In all these process, the LRR domains are mainly involved in either protein-protein interactions or specific receptor-ligand binding.
