Changes of cholinergic nerve morphosis and density in the left ventricle of rats after acute ischemic injury induced by myocardial infarction

摘要 目的观察大鼠心肌缺血性损伤后,左心室胆碱能神经纤维形态及密度的变化。方法将90只 Wistar大鼠分为4组(A组15只,B、C、D组各25只)。A组为假手术组,实验组(B、C、D)分别结扎左冠状动脉前降支(LAD)形成急性前壁心肌梗死(AMI),梗塞后取材时间分别为6 h(B组)、12 h(C组)、48 h(D组)。应用 Karnovsky-Roots法,显示梗塞后不同时间左心室心肌胆碱能神经纤维的分布。结果 A组大鼠左心室心肌各层均见深褐色的胆碱能神经纤维沿肌间走行,B、C、D各组胆碱能神经纤维呈断裂、散在分布。A组左心室前壁胆碱能神经纤维密度为(1.52±0.16)%,B组为(1.28±0.15)%、C组为(1.02±0.11)%、D组为(0.65± 0.09)%,B、C、D各组与A组比较,差异均有统计学意义(P<0.05、P<0.01、P<0.01)。各组左心室后壁胆碱能神经纤维密度无明显变化。结论大鼠左心室前壁心肌缺血性损伤及坏死后,左心室前壁胆碱能神经呈断裂状,神经密度随时间延长而降低。 Objective To observe morphosis and density of cholinergic nerves in the left ventricule of heart after acute ischemic injury. Methods 90 Wistar rats were used in this study. The rats were randomly divided into 4 groups. Group A was normal control group. Left anterior descending coronary artery of rats in experiment groups was ligated to form acute myocardial infarction (AMI). Hearts were excised 6 hours (group B), 12 hours (group C)and 48 hours (group D) after AMI. Karnovsky-Roots methods of histochemical techniques were used to show the distribution of cholinergic nerves. The density of cholinergic nerves was calculated by Medical Image Analysis Processing System. Differences in density among groups were compared by t-test. Results The cholinergic nerves went along the myocardium in group A. The cholinergic nerves were disrupted in the B,C and D groups .The cholinergic nerves density decreased in the left ventricular anterior wall:being (1.52 ± 0.16)% in group A, (1.28 ± 0.15)% in group B, (1.02 ± 0.11)% in group C, (0.65 ± 0.09)% in group D. The nerve densities in groups B,C and D were respectively compared with that in the group A, the differences were statistically significant (P 〈 0.05,P 〈 0.01 ,P 〈 0.01 ). However, the cholinergic nerves density was normal in the posterior wall. Conclusions The cholinergic nerve density decreased in the left ventricule of heart after AMI.
