Determining fluoride of micro-quantity in serum using diffusion-electrode method

摘要 目的建立一种新的微量血清氟测定方法。方法应用微扩散-电极法测定血清氟,并通过不同种类、浓度扩散酸,不同扩散时间等因素评价该测定方法的可行性。结果将1.00 ml血清样品加2 ml扩散酸 (AgSO4-H2SO4-HMDS)在室温下扩散15-20 h,绘制标准曲线各浓度点的变异(CV)均小于5%,相关系数(|r|) 平均为0.999 2,a=109.66,b=56.97,标准曲线的线性回归及电极斜率均符合要求。方法的精密度:当血清氟为0.035、0.098、0.49 mg/L时,CV分别为8.00%、5.51%、2.42%;准确度:用该法测定血清标准物质含氟量,分别为(1.03±0.06)mg/L和(5.07±0.13)mg/L时,其结果与给定值的相对偏差分别为4.85%、1.58%。当血清氟本底值为0.035、0.082、0.215 mg/L,加氟标量分别与血含氟量相当,其平均回收率分别为(95.0±7.24)%、(96.8± 5.35)%、(96_3±4.76)%。结论应用该法测定微量血清氟准确度、精密度均符合生物样品分析要求。 Objective To set up a new method for determining fluoride of micro-quantity in serum. Methods Fluoride content was determined using diffusion-electrode method under different diffusion conditions, such as different types and concentrations of diffusion-acid and different diffusion time. The feasibility of the method was thus evaluated. Results 1.00 ml serum added up with 2 ml diffusion acid(AgSO4-H2SO4-HMDS) was diffused at the room temperature for 15 - 20 h. The serum fluoride was then determined with the electrode. The CV(%) of various concentration of the standard curve was all lower than 5%. The average of the correlation coefficient (│r│) was 0.999 2, a = 109.66, b = 56.97 (slope). The linear correlation of the standard curve and the slope of the electrode met the demands. When serum fluoride values were 0.035, 0.098, 0.49 mg/L, variation coefficients were 8.00%, 5.51%, 2.42% respectively. Accuracy, when the standard serum fluoride values being (1.03 ± 0.06)mg/L and (5.07 ± 0.13)mg/L were determined, relative deviation were 4.85%, 1.58% respectively. When the background values of serum fluoride were 0.035, 0.082, 0.215 mg/L, rates of recovery were (95.0 ± 17.24)%, (96.8 ± 5.34)%, (96.3 ± 4.76)%. Conclusions Accurate and precise in determining fluoride of micro-quantity in serum, this method meets the demands of analyses bio-samples.
