摘要
目的探讨托吡酯(TPM)对癫大鼠血清瘦素水平和下丘脑弓状核神经肽Y(NPY)表达的影响及两者与体质量变化的关系。方法115只大鼠随机分成分别为空白组、TPM对照组、模型组和TPM治疗组。称重1次/3d,分别于2、4、6周末用放射免疫法测血清瘦素水平,用免疫组织化学染色法测下丘脑弓状核NPY表达。结果TPM对照组和TPM治疗组实验第4周出现体质量下降(P<0.05),同时在第2、4周末两组血清瘦素较空白组、模型组明显升高(P<0.05),且随时间下降。TPM治疗组与TPM对照组大鼠NPY神经元表达较空白组和模型组升高(P<0.05),且随时间渐上升。结论TPM能提高大鼠血清瘦素水平,从而降低体质量,但瘦素升高使体质量下降与NPY无关。
Objective To study the effect of topiramate (TPM) on sertum level of leptin and the expression of neuropeptide Y (NPY) in arcuate nucleus and their relationship with weight changes in pentetruzole-induced epileptic rats, and to explore the mechanism that TPM could reduce the weight. Methods One hundred and fifteen Wistar rats were randomly divided into 4 groups: blank control group,TPM contrast group, pentylenetetrazo[(PTZ) model group, and TPM group. Weight changes were measured every 3 days. Serum leptin level was detected with rodioimmunity method and hypothalamic areuate nucleus NPY expression was measoured by immunohistochernical ABC technique at the 2^rd, 4^th, 6^th weekend, respectively. Results in TPM control group and TPM group weight loss appeared in the 4^th week compared with other groups at the 3^rd weekend and fell to the lowest at 4^th weekend ( P〈 0.05). Serum leptin level in the 2 groups rose clearly at the 2^rd and 4^th weekend compared with blank group and PTZ group and fell to normally level at the 6^th(P〈0.05). it descended gradually during the period. Expression of NPY immvm-neurons in both groups grew obviously compared with blank group and PTZ group, Conclusion TPM can reduce the rats' weight through raising the level of serum leptin, but the rat weight loss with the raise of leptin levels was not by suppressing the expression of NPY.
